Is this supported?  Is it simply just a case of opening up the proper ports on our corporate firewall for SpyraTest to to talk to our internal Jira server?

Yes this is supported. You just need to use the Desktop DataSync tool to connect to SpiraTest and JIRA. It connects to both using web services over port 80/443 so as long as the computer running Desktop DataSync can see both our hosted server and your internal JIRA instance, no firewall changes should be necessary.

Where do you configure the Jira server info for the Desktop DataSync tool?

You configure that inside SpiraTest itself in the Administration > Data Synchronization section.
